چکیده Abstract: COVID-19 individuals with varied severity groups, including moderate, mild, also severe, are affected via the cytokine storm brought on via SARS-CoV2 infection, It is a major contributor to acute respiratory distress syndrome (ARDS). The current research 's goal was to examine IFN-Ɣ gene expression in COVID-19 at various severity levels.Methods: The research include 140 divided as 105 COVID-19 positive patients (46 males also 59 females) also 35 COVID-19 negative healthy peoples (16 male also 19 females) (control). The separation of Peripheral blood mononuclear cells (PBMC) was achieved using Ficoll also then RNA were extracted converted to cDNA also the gene expression using GAPDH as housekeeping gene. Results: The research include 105 COVID-19 patients (35 for each Mild, moderate also sever groups) also 35 healthy control (COVID-19 negative). The findings found no statistically significant changes. at p < 0.05 in age among different COVID-19 groups also control (f-ratio value is 0.542 also p-value is 0.653). The finding of gene expression reveal over expression of IFN-Ɣ gene among COVID-19 patients when compared with health control (non-COVID-19) also the relative quantification (fold change) (Mean±SD ) were 8.60±9.46, 6.424±1.03, 7.887±1.88, also 11.510±15.98 for All, Mild, Moderate also Sever COVID-19 patients respectively. The finding of one way ANOVA test RQ (Fold change) IFN-Ɣ (Mean±SD) for Mild, moderate also sever groups revealed non-significant at p < 0.05, the f-ratio value is 2.76803 also the p-value is 0.067507. Conclusions: The present research conclude upregulation of IFN-γ gene in PBMC of COVID-19 positive patients without significant differences among different severity groups.
