Introduction: Neuroinflammation plays a key role in the pathogenic mechanisms related to cerebral ischemia. Ischemic neurons are activated for inducible nitric oxide synthase (iNOS) during the early stages of reperfusion after ischemia. Inflammatory cells begin to produce large amounts of inflammatory proteins, including the pathological and inducible form of NOS, which causes nitrosative stress that leads to further brain damage. The beneficial effects of exercise have shown that exercise preconditioning can have neuroprotective effects after cerebral ischemia. However, the role of physical exercise in the expression of iNOS induced by cerebral ischemia has not yet been fully elucidated. The aim of the present study was to investigate the effect of sports activity before cerebral ischemia on the expression of iNOS. Materials and methods: Rats were divided into three groups, sham group, training+ischemia group (4 weeks of training before induction of ischemia) and ischemia group (7 rats in each group). In the ischemia group, animals underwent occlusion of both common carotid arteries (CCA). In the exercise+ischemia group, animals exercised for 4 weeks before induction of ischemia. Cerebral ischemia model was established by occlusion of common carotid arteries. Evaluation of iNOS protein expression level in the hippocampus area was done by ELISA method. Results: In biochemical analysis, the level of hippocampal iNOS protein expression was higher in the ischemia group than in the sham group. Pre-ischemic exercise decreased the level of iNOS compared to the ischemia group. Conclusion: The findings of this study showed the neuroprotective potential of sports activity through the reduction of iNOS in the rat model of cerebral ischemia.
