EVIDENCE FOR AMELIORATIVE EFFECT AND MECHANISM OF ALPHA-TOCOPHEROL SUPPLEMENTATION AGAINST D-GALACTOSE-INDUCED TESTICULAR ALTERATIONS IN RATS: HISTOLOGICAL AND IMMUNOHISTOCHEMICAL STUDIES.

Abstract

There is a growing interest in exploring the relation between aging and male infertility, as well as developing anti-aging interventions. Alpha-tocopherol (α-T) has been effectively used as an antioxidant molecule in various tissues. However, the effect of α-T on D-galactose (D-Gal)-induced testicular aging have not been previously examined. Consequently, this study aimed to evaluate the ameliorative potential of α-T against D-Gal-induced testicular changes. Twenty-eight male Wistar rats were randomly allotted into four groups (n=7). Group I (Control, received intraperitoneal injection of PBS), Group II (200 mg/kg body weight D-Gal, intraperitoneally), Group III (200 mg/kg body weight of D-Gal with 200 mg/kg body weight of α-T), Group V (received 200 mg/kg body weight of α-T). At eight weeks of treatments, blood samples and tissues were collected for measuring testosterone, oxidative stress parameters and conducting Hematoxylin and Eosin staining. Also, immunohistochemical method for CYP19 and 17β-HSD-3 expression of testicular tissue marker was applied. The findings revealed that the injection of D-gal impaired the immunoreactivity of CYP19 and 3β- HSD, but α-T reversed the decrease in the expression of these enzymes. The tubular diameter, the epithelial height, and the number of Leydig cells were significantly increased in D-Gal + α-T group compared to DGal- treated rats. In addition, α-T co-exposure caused an increase in testosterone as well as SOD concentrations and also a decrease in MDA level. All in all, the results of this study pointed out that α-T had an ameliorative effect against D-Gal-influenced testis alterations via restoring histoarchitecture, reversing oxidative and steroidogenesis parameters, and stabilizing testosterone hormones
